Adapting fish defenses to block human infections: Antimicrobial peptide of fish gills inspire clean surfaces

ScienceDaily (Nov. 29, 2012) ? An undergraduate research team analyzes peptides from fish gills to engineer antimicrobial surfaces for food preparation and medical devices.

Living in an environment teaming with bacteria and fungi, fish have evolved powerful defenses against waterborne pathogens, including antimicrobial peptides located in their gills. Undergraduate researchers at Worcester Polytechnic Institute (WPI) are studying the biology and the mechanics of one of those peptides with the hope they can use that knowledge to create engineered surfaces that kill bacteria responsible for foodborne illnesses and hospital-acquired infections.

The research team, led by Terri Camesano, professor of chemical engineering, reports its latest findings in the paper "Creating Antibacterial Surfaces with the Peptide Chrysophsin-1," published online in October by the journal ACS Applied Materials & Interfaces.

"Fish have a wonderful solution for blocking bacterial and fungal infections," Camesano said. "In this study, we are working to better understand the biochemical mechanics of that process."

As fish filter water through their gills to extract oxygen, antimicrobial peptides (AMPs), including Chrysosphin-1, trap and kill pathogens before they can invade the fish's bloodstream. Scientists in many laboratories around the world are actively exploring the potential use of these molecules to prevent human infections. In the current study, the WPI team attached AMPs to silicon and gold surfaces using two different approaches and measured the bound peptides' ability to kill the bacterial pathogen E. coli.

In the first method, the AMPs were absorbed directly onto gold and silicon crystals, forming a single layer of molecules with the AMPs lying flat on the surface. In the second method, the tips of the AMPs were attached to the surfaces with a glue-like substance so that the peptides rose vertically, like blades of grass extending up from the ground. Surfaces with both AMP configurations were cultured with E. coli cells. The results showed that when the AMPs were lying flat they killed 34 percent of the bacteria in the culture, but when they were standing up vertically they killed 82 percent.

"The hypothesis is that when peptides are attached vertically to the surfaces, they are better able to move and bend so they take on a shape that is more effective in binding to and disrupting the E.coli cells," Camesano said.

In addition to gathering data about the antibacterial efficacy of the attached AMPs, the WPI research team developed a technique for monitoring, in real time, the attachment of AMPs to surfaces. Using quartz crystal microbalance with dissipation monitoring (QCM-D), the team measured the quantity of AMPs that successfully attached to the surfaces in the horizontal and vertical orientations and the density of the AMP layers, along with other properties.

"This was a powerful process, to be able to essentially watch the binding process as it happened," Camesano said. "It is a technique that we will continue to apply in further studies."

Camesano said gold and silicon surfaces were selected for the current study because their chemical properties are well-suited for AMP binding. In ongoing work, Camesano's laboratory will continue to characterize the mechanics of AMP binding for optimal antimicrobial activity and test other materials, including titanium, stainless steel, and plastics, that would have greater utility in food preparation and healthcare.

"What is also notable about this study is that it is the work of undergraduates," Camesano said. "They've done excellent work here that will inform future graduate studies in our lab."

The WPI undergraduates who co-authored the AMP paper are Ivan Ivanov '12 (chemical engineering), Alec Morrison '12 (biochemistry), and Jesse Cobb '12 (chemical engineering). Co-author Catherine Fahey, a student at George Washington University, worked on the study during at a summer project at WPI. Their work was sponsored in large part by a grant from the National Science Foundation's Research Experiences for Undergraduates (REU) program.

Duke Energy CEO to leave in NC merger settlement

RALEIGH, N.C. (AP) ? Duke Energy CEO Jim Rogers will step down as head of the largest U.S. electric utility by the end of 2013 as part of a settlement with the North Carolina utilities regulator that ends an investigation into the company's takeover of in-state rival Progress Energy.

The North Carolina Utilities Commission and Duke Energy said Thursday the deal concludes the regulator's probe into whether the company misled the commission ahead of the merger approval. The commission had the power to reverse or alter its approval.

Hours after the merger was completed July 2, Duke Energy's board ousted Progress Energy CEO Bill Johnson, who was supposed to take over the combined company. It had promised to keep him in place throughout the 18-month process of merging the two Fortune 500 energy companies headquartered in North Carolina. The deal created the nation's largest electric company.

The state regulator and Attorney General Roy Cooper launched investigations that demanded several internal Duke documents and communications. The commission hired a former federal prosecutor to probe whether Duke Energy executives and board members were telling regulators that Johnson would head the combined company while secretly arranging to dump him. A Cooper spokeswoman said the attorney general's investigation will continue.

Johnson was hired earlier this month as chief executive of the Tennessee Valley Authority, the nation's largest public utility.

"This settlement agreement is an important step forward for the company because it resolves one of our key near-term priorities: bringing closure" to the commission's review, Rogers said in a statement. His retirement will take effect by Dec. 31, 2013.

Duke Energy now has 7.1 million residential and business customers in North Carolina, South Carolina, Ohio, Kentucky, Indiana and Florida. It did not name a replacement CEO immediately.

Duke Energy also required the regulator's approval to raise electricity rates for its 1.9 million North Carolina customers, a request the Charlotte-based company plans to file as early as February.

Rogers became president and CEO of Duke Energy after its 2006 merger with Cincinnati, Ohio-based Cinergy, which he had headed for 11 years. Rogers had planned to retire as Duke's CEO at the end of this year before the shakeup with Johnson.

When he took over as head of the combined companies, now called Duke Energy, his departure date was no longer set.

The commission is scheduled to stamp its approval on the deal on Monday.

While the settlement specifies several management comings and goings, it does not involve the regulator in the company's decision-making, as some of the company's supporters had feared.

Chief Legal Officer Marc Manly will give way to a new top lawyer by the end of the year. Lloyd Yates, who moved from Progress Energy, will take over as executive vice president responsible for its regulated state power subsidiaries. Former Progress Energy General Counsel John McArthur, who resigned after the takeover, will get a two-year position advising on regulatory and legislative matters in North Carolina.

Duke Energy agreed to stock the committee searching for Rogers' successor with a balanced number of former Duke and former Progress board members, plus a new outside board member. The committee also will search for two new board members to replace former Progress directors who resigned in protest this summer.

A new board committee will meet with the utilities commission to hear what they think about the board's activities.

Duke Energy promised to pass along to North Carolina customers rather than shareholders an extra $25 million in merger-related savings, spend another $5 million on workforce development and low-income assistance, and promise to keep at least 1,000 employees in Raleigh for at least 5 years.

While Duke Energy denied wrongdoing, the utilities commission said the settlement includes the company issuing a statement acknowledging it has "fallen short of the commission's understanding of Duke's obligations" as a regulated utility.

An issue I feel strongly about is using scores from standardized tests to determine (assess) an individual?s speech and language abilities. I learned about what validity and reliability mean in reference to using standardized testing tools early in my educational course work for this field. What is bothersome to me is that while it is common knowledge that most if not all exams, especially those for children, are not ?normed? on children I would test, many agencies and supervisors demand a score as part of an evaluation report.

Children I would test are those that live in New York City and the New York region (i.e., Long Island, Yonkers, Westchester). Further exasperating this topic are those children which are exposed to more than one language. These children do not follow the ?typical? speech and language developmental norms of any monolingual community. Therefore, why are SLPs using these testing tools to make an assessment? Some may think the SLP is monolingual and may not be aware. Let?s make it clear, there is no SLP that should graduate from any program, bilingual or not, that is not aware of reading a testing manual at the least. We are also taught, even if briefly, that testing tools should be scanned to remove or adjust bias questions/materials and that bilingual children should be tested by bilingual SLPs. Let me make another point clear: while a person is bilingual it does not qualify them to assess a bilingual person. For example, I am proficiently bilingual in English and Spanish. ??

What is especially worrisome is that while this is such a touchy subject, a certified SLP can actually lose their license over this issue/topic (using a score to report findings). I would venture and guess that one or two malefactions would not substantiate a license being revoked, however it baffles me as to how not more of an emphasis is made on using appropriate testing materials and informed clinical opinions to report findings instead of using scores from a test. In New York City there are rarely any children that would meet the reliability, validity, and norming standards for any test that I know of, yet I find that too often they are used to assess the child?s language abilities.

Catherine J. Crowley is not only the director of the Bilingual Institute at Teachers College, Columbia University but she is an advocate for this issue. In reality, I cannot say she is an advocate she is the leader who identifies what issues need to change and how to go about changing them. Last year when I started at the institute she informed us of some upcoming trainings and reforms regarding this precise topic: agencies forcing their employees to use scores to report a disorder/delay. Cate, as she likes to be addressed by her students, is still conducting trainings around the city to various agencies and changing policies as we speak. She has a lot of support, some of which is through her students and staff. Cate posts information on the institution?s Facebook page such as rallies, petitions, current events, etc. This is a person that I would reach out to about this issue and support for continuing to proctor this change.?

Schapiro stepping down at SEC, Walter to step in

WASHINGTON (Reuters) - The head of the U.S. Securities and Exchange Commission, Mary Schapiro, will step down next month after a tumultuous four years spent rehabilitating the agency's battered reputation, handing the reins temporarily to a close ally.

SEC Commissioner Elisse Walter, a career regulator who has sided with Schapiro on most of the critical issues before the agency, was named chairman-designate and could serve until December 2013, buying time for President Barack Obama to win Senate approval for a long-term replacement.

Obama plans to nominate someone soon, a White House official said. Walter is among the candidates likely to be considered, as is Treasury official Mary Miller, who spent nearly three decades at T. Rowe Price and has been outspoken about the need to make money markets safer for investors.

The new SEC chair will need to finish Schapiro's task of resurrecting the agency's reputation, which was badly tarnished by the 2007-2009 financial crisis.

The SEC is considering money market reforms, additional market structure safeguards, and still needs to write a number of major rules dictated by the 2010 Dodd-Frank financial reform law, including a final version of the Volcker rule to ban banks from trading for their own accounts.

"Elisse has been viewed as being in tune with Mary Schapiro's agenda," said Barry Barbash, a former director of the SEC's division of investment management, now an asset management lawyer at Willkie Farr & Gallagher LLP in Washington. "If the idea is to keep the SEC running the way it's been running with the same policies, Elisse would seem an appropriate choice."

Speculation has swirled for months that Schapiro would leave soon after the November 6 presidential election.

Her departure leaves the commission split 2-2 between Democrats and Republicans, which could make it harder for the commission to come to agreement. Obama could move to appoint a fifth commissioner before naming a new chairman.

When Schapiro took over in 2009, the agency was under heavy fire for regulatory blindspots that critics said helped fuel the crisis. It was also lambasted for failing to catch now-convicted Ponzi schemer Bernard Madoff, whose fraud cost investors an estimated $65 billion.

In addition to shoring up the agency's name, Schapiro had to fight numerous other fires - from the 2010 "flash crash" that sent the Dow Jones industrial average tumbling 700 points within minutes to high-profile court losses.

"We've gotten a lot done, I'm really proud of where the agency is today, so it seemed like a good time," Schapiro said in an interview on Monday after announcing her departure.

UNFINISHED BUSINESS

Other possible replacements for Schapiro include Sallie Krawcheck, a former top executive at Bank of America and Citigroup, and Richard Ketchum, chairman of FINRA, Wall Street's self-funded regulator. SEC enforcement director Robert Khuzami, a Republican, is considered a long shot.

While Walter is also a possibility, she has already been at the SEC for more than four years and served as acting chairman before Schapiro's confirmation.

In many ways, Walter and Schapiro have been joined at the hip in their career experience and orientations. They both spent years as attorneys at the Commodity Futures Trading Commission and at the SEC, with stints in top positions at FINRA.

"She is a strong advocate of disclosure, of regulation and of consumer protection," former SEC Commissioner Edward Fleischman said of Walter.

SCHAPIRO's LEGACY

Schapiro's time at the SEC was marked by some controversy and her departure leaves uncertainty around major initiatives. But former SEC officials said Schapiro helped revive a moribund agency.

"I think she saved the SEC, which was close to extinction when she took over," former SEC Chairman Arthur Levitt told Reuters.

Schapiro said in the interview that steering the agency out of that period in its history was one of the highlights of her tenure there.

She streamlined the SEC enforcement process, hired new types of employees and created a new tips database and a whistleblower office. In the past two years, the agency logged a record number of enforcement actions and brought major financial crisis cases, including a record $550 million settlement in 2010 with Goldman Sachs.

It has also implemented reforms to protect markets against major swings caused by errant technology - as was the case with the flash crash.

"The SEC is stronger, and our financial system is safer and better able to serve the American people - thanks in large part to Mary's hard work," Obama said in a statement.

But the agency has also been bogged down with major rules the 2010 Dodd-Frank financial regulation law required it to write, many of which are still in process.

Business groups have challenged much of the SEC's recent rule-making efforts and won major battles, including convincing a federal appeals court to throw out the agency's "proxy access" rule, which would have empowered shareholders to nominate directors to corporate boards.

Schapiro did not say what she planned to do next.

Ranbaxy recalls generic Lipitor doses

(AP) ? Ranbaxy Pharmaceuticals Inc. has recalled dozens of lots of its generic version of cholesterol drug Lipitor because some may contain tiny glass particles, the latest in a string of manufacturing deficiencies that once led U.S. regulators to bar imports of the Indian company's medicines.

Ranbaxy, a subsidiary of Ranbaxy Laboratories Ltd., India's biggest drugmaker, is operating under increased scrutiny from the U.S. Food and Drug Administration because of quality lapses at multiple Ranbaxy factories over the past several years. The FDA also has alleged the company lied about test results for more than two dozen of its generic drugs several years ago.

On Friday, Ranbaxy posted a notice on its U.S. website, saying it's recalling 10-, 20- and 40-milligram doses of tablets of atorvastatin calcium. That's generic Lipitor, the cholesterol fighter that reigned for years as the world's top-selling drug.

The recall includes 41 lots of the drug, nearly all with 90 pills per bottle, but three lots contain 500 pills per bottle. It's unclear how many bottles are in each lot, but medicine batches typically contain many thousands of pills. The 80-milligram strength tablets are not affected.

Ranbaxy spokesman Chuck Caprariello did not answer questions or provide any additional information beyond the statement on the company's website.

"Ranbaxy is proactively recalling the drug product lots out of an abundance of caution," the website statement read. "This recall is being conducted with the full knowledge of the U.S. FDA."

The company also filed a two-sentence statement with the Bombay Stock Exchange stating Ranbaxy's investigation would be completed within two weeks, but that after that temporary disruption to the U.S. supply, the company expected to resume shipments here.

Patients who've filled a prescription can contact their pharmacy to determine whether it was made by Ranbaxy or another generic drugmaker and, if it's from Ranbaxy, whether it came from a recalled lot.

Ranbaxy's manufacturing deficiencies, dating to 2006, led to a lengthy investigation and sanctions by the FDA. During the probe, federal investigators found Ranbaxy didn't properly test the shelf life and other safety factors of its drugs and then lied about the results.

In mid-2008, the FDA barred Ranbaxy from shipping into the U.S more than 30 different drugs made at factories in India. Meanwhile, the U.S. Department of Justice demanded Ranbaxy turn over internal documents, alleging the company lied about ingredients and formulations of some medications.

In early 2009, the FDA said it would not consider any new applications from Ranbaxy to sell in the U.S. any products made at the troubled factories.

As FDA discussions with Ranbaxy continued, it appeared Ranbaxy would lose its shot at a revenue windfall when Lipitor's generic U.S. patent expired last Nov. 30. At the time, Lipitor brought in almost $8 billion a year in U.S. sales.

As often happens when patents first expire, for the first six months only one generic rival could compete with brand-name Lipitor. Ranbaxy had that right, although an authorized generic from Lipitor maker Pfizer Inc. and partner Watson Pharmaceuticals Inc. went on sale on Dec. 1. With competition so limited, the generic prices only declined a bit from brand-name drug's price of about $115 a month ? until several other generics entered the market six months later.

The FDA finally ended the suspense, deciding just before midnight on Nov. 30 to let Ranbaxy sell generic Lipitor made at the company's Ohm Laboratories factory in central New Jersey. It was unclear Friday whether the recalled Ranbaxy pills were made there or elsewhere.

Meanwhile, Ranbaxy is operating under a settlement with the FDA, called a consent decree, signed on Dec. 20, 2011. It requires Ranbaxy to improve manufacturing procedures, ensure data on its products is accurate and undergo extra oversight and review by an independent third party for five years. Ranbaxy at the time set aside $500 million to cover potential criminal and civil liability stemming from the Justice Department investigation.
